After a single dose, tadalafil typically stays in the body for about 2 to 3 days (48 to 72 hours) in noticeable amounts. Its half-life is around 17.5 hours, meaning it takes that long for the concentration in the blood to reduce by half. Because of this slow elimination, small traces can remain for up to 4–5 days, especially in people with slower metabolism, older age, or kidney/liver issues.

The main functional effect (erection support and mild blood pressure lowering) usually lasts around 24 to 36 hours, but the strongest effect is within the first 2 to 12 hours after taking it. After that, the effect gradually reduces while the drug is still being cleared from the system.
